Which protocol should I follow for DNA extraction from plant tissue?
Bionano Inc. offers 4 protocols for DNA extraction from plant tissue. From our humble experience based on DNA extraction in soybean, strawberry and maize, we recommend the use of Bionano Prep Plant Tissue DNA Isolation High Polysaccharide Protocol (30168).
What kind of plant tissue should I use for DNA extraction?
Fresh folded young leaves are highly recommended. They contain high concentration of DNA and low amount of polysaccharide.
Do I need to perform dark treatment for plant sample?
Dark treatment is not necessary for folded young leaves. However, if you use unfolded young leaves for DNA extraction, 24 to 48 hours dark treatment should be applied.
How many plugs can be prepared from 0.5 g plant tissues?
Usually up to 5 plugs are prepared. In general, the concentration of plugs (5 plugs) prepared is 50-130 ng/ul (The suggested concentration of plug is 35-200 ng/μl). If you observe large pellet with high DNA content, more plugs can be prepared by titrating schemes.
